What is albumin-bound paclitaxel? Albumin-bound paclitaxel (commonly known as albumin paclitaxel, also abbreviated as Nab-P) is a new paclitaxel nanoformulation, which is internationally recognized as the most advanced formulation of paclitaxel. It combines endogenous human albumin with paclitaxel in a non-covalent form and the resulting nanoparticles completely overcome the disadvantage of paclitaxel being insoluble in water, without the need for polyoxyethylene castor oil or Tween 80 as a co-solvent. As albumin is an endogenous natural product, it has the advantages of being safe, non-toxic, non-immunogenic, biodegradable, and biocompatible.
Why is albumin-bound paclitaxel the most advanced?
1. No co-solubilizationno pre-treatment
Adopting plasma protein of natural origin, it binds hydrophobic paclitaxel molecules in a non-covalent form without any co-solvent, eliminating the hypersensitivity and toxicity reactions caused by co-solvents, thus eliminating the need for hormone pretreatment before use and shortening the infusion time to 30 minutes, greatly shortening the hospital stay of oncology patients, and even completing chemotherapy through day clinic.
2. Better pharmacokinetics
Compared with conventional paclitaxel, albumin paclitaxel has a faster and higher tissue distribution in the body, as well as slower excretion from the tissues, the tolerated dose in the human body can be significantly increased and the therapeutic effect is significantly improved.
3. Selective local tumor aggregation
One of the reasons why many patients are afraid of paclitaxel chemotherapy is that the distribution of traditional paclitaxel in tumor tissues and other normal tissues is basically the same, causing many serious side effects. In contrast, paclitaxel has a miraculous “selective local tumor enrichment” and its concentration in tumour tissue is much higher than that in other normal tissues.
